Abstract:

Background: In an ectopic pregnancy (EP) the egg does not reach to the uterus, but instead implants somewhere outside it, usually in fallopian tube. In most cases the exact cause of an ectopic pregnancy is unknown but it is often a result of some sort of damage to the fallopian tube. The tube may have become blocked or narrowed by previous surgery or infection. An active immune system need to react against these infections. Toll-like receptors (TLRs) are a family of innate immunity receptors that are essential for detecting pathogens. Different TLRs serve as receptors for diverse ligands, for example: TLR4 is the receptor for Gram-negative lipopolysaccharide (LPS) and it show higher expression in the Fallopian tube and endometrium compared with that in the cervix. The objective of this study is to clarify whether TLR4 expression level is involved in EP or not? Materials and Methods: In this study, our samples were divided into two groups: case and control groups. Case group were women underwent salpingectomy for EP and control group was pseudo-pregnant women with healthy tube underwent hysterectomy. For control group, Women were injected with human chorionic gonadotropin (hCG) in 14 days leading up to hysterectomy to produce a state of pseudo-pregnancy. In this investigation TLR4 expression level was detected in infundibulum, ampulla and isthmus of tubes in case and control groups and also Q-PCR was used to compare quantitative expression of TLR4 gene among case and control groups. Results: Using RT-PCR shown that TLR4 gene was expressed in all region of control group, but in case group was not expressed in infundibulum and ampulla, whiles very, very low expression in isthmus was detected. This finding was confirmed by Q-PCR for TLR4 gene. Conclusion: No expression of TLR4 in fallopian tube with EP is a strong evidence of critical role of this receptor in prevention of ectopic pregnancy.

P-83: Immunological Evaluation of Ectopic Pregnancy with Emphasis on TLR5

Background: The presence of immune system within the female reproductive tract is increasingly being recognized as an important element for maintaining host protection and promoting reproductive functions. The Toll-Like Receptors (TLRs) family plays an important role in innate immune responses against microbial pathogens within the genital tract. These receptors recognize distinct pathogen-asso...

P-246: Gene Variation of TLR4 in Patients with Endometriosis

Background: Endometriosis, defined as the presence of endometrial tissue outside of the uterus, is an estrogen-dependent chronic inflammatory condition associated with degrees of pelvic pain and infertility. Toll-like receptors play a key role in immune response, by regulating inflammatory reactions and activating adaptive immune response to eliminate infectious pathogens and cancer debris. Pol...

TLR4 links podocytes with the innate immune system to mediate glomerular injury.

Toll-like receptors (TLR) classically recognize pathogen-associated danger signals but are also activated via endogenous ligands. For evaluation of their role in inflammatory kidney disease, the function of TLR was analyzed in two mouse models of cryoglobulinemic membranoproliferative glomerulonephritis (MPGN; mice transgenic for thymic stromal lymphopoietin [TSLP], with or without deletion of ...

A comprehensive review on vitamin D receptor (VDR) gene polymorphism in immune-related diseases with emphasis on dermatologic disorders

There are many immune mediated disorders with the corroborated role of vitamin D or Vitamin D Receptor (VDR) gene polymorphisms in their pathogenesis, immunologic regulation, and disease characteristics. Therefore, in this review, we searched PubMed data base in regard to the role of VDR gene polymorphisms in common autoimmune disorders, emphasizing on dermatologic diseases.

Pulmonary Metastatic Choriocarcinoma in a Patient with Ectopic Pregnancy

Gestational trophoblastic tumor (GTT) most commonly follows a molar pregnancy. In fact, it can occur following gestational events such as induced or spontaneous abortion, ectopic pregnancy, and term pregnancy. In this study, we present the case of a patient with ectopic pregnancy who was treated with a single dose of methotrexate (MTX). The maximum titer of beta subunit of human chorionic gonad...
